A bioprocess is a specific process that uses complete living cells or their components e. Transport of energy and mass is fundamental to many biological and environmental processes. Areas, from food processing including brewing beer [1] to thermal design of buildings to biomedical devices to pollution control and global warming , require knowledge of how energy and mass can be transported through materials momentum, heat transfer, etc.

Addressing the Challenges in Downstream Processing Today and Tomorrow

The production of biopharmaceuticals, known as bioprocess, involves a wide range of techniques. In this review, we discuss the technology involved in the bioprocess and describe the available strategies and main advances in microbial fermentation and purification process to obtain biopharmaceuticals. Biopharmaceuticals are mostly therapeutic recombinant proteins obtained by biotechnological processes. They are derived from biological sources such as organs and tissues, microorganisms, animal fluids, or genetically modified cells and organisms. This investment is justified by the well-characterized genomes, versatility of plasmid vectors, availability of different host strains, cost-effectiveness as compared with other expression systems.

Downstream processing of viral vectors and vaccines

Downstream processing refers to the recovery and the purification of biosynthetic products, particularly pharmaceuticals, from natural sources such as animal or plant tissue or fermentation broth, including the recycling of salvageable components and the proper treatment and disposal of waste. It is an essential step in the manufacture of pharmaceuticals such as antibiotics, hormones e. Downstream processing is usually considered a specialized field in biochemical engineering , itself a specialization within chemical engineering , though many of the key technologies were developed by chemists and biologists for laboratory-scale separation of biological products. Downstream processing and analytical bioseparation both refer to the separation or purification of biological products, but at different scales of operation and for different purposes. Downstream processing implies manufacture of a purified product fit for a specific use, generally in marketable quantities, while analytical bioseparation refers to purification for the sole purpose of measuring a component or components of a mixture, and may deal with sample sizes as small as a single cell. A widely recognized heuristic for categorizing downstream processing operations divides them into four groups which are applied in order to bring a product from its natural state as a component of a tissue, cell or fermentation broth through progressive improvements in purity and concentration.

Downstream processing in biotechnology concerns the separation and purification of the bio-product downstream of the fermenter as the name implies and this is the focus of this book.

In recent years, most pharmaceutical companies have focused on the development of monoclonal antibodies mAbs. Increasing upstream titers and shrinking development timelines have posed several challenges to downstream process development of mAbs. Some of the major strategies and tools to address these challenges include the development of highly efficient platforms, high-throughput screening HTS tools, and reduction of the number of unit operations to help with facility fit. In the future, mAbs may represent a smaller percentage of the pipeline as portfolios concentrate more on development of antibody fragments, nanobodies, biosimilar protein therapeutics, conjugated proteins and vaccines, Fc-fusions, and nonantibody protein scaffolds. In addition to high cell density mammalian expression, expanded utilization of other expression systems such as microbial and yeast will support these newer biotherapeutics BioTx. The diversity of BioTx and expression systems will pose unique challenges for downstream development. Early biotechnology products in the US and Europe were characterized by low titers and low cell densities using various cell hosts 1, 2.
