Protein purification is a series of processes intended to isolate one or a few proteins from a complex mixture, usually cells , tissues or whole organisms.

NCBI Bookshelf. An adage of biochemistry is, Never waste pure thoughts on an impure protein. Starting from pure proteins, we can determine amino acid sequences and evolutionary relationships between proteins in diverse organisms and we can investigate a protein's biochemical function. Moreover, crystals of the protein may be grown from pure protein, and from such crystals we can obtain x-ray data that will provide us with a picture of the protein's tertiary structure—the actual functional unit. Purification should yield a sample of protein containing only one type of molecule, the protein in which the biochemist is interested.

Separation techniques: Chromatography

Figure 1: Stain-free gels are more sensitive than Coomassie-stained gels. TGX stain-free gels BioRad Laboratories were loaded with standards and a serial dilution of a protein mixture with four different proteins of varying tryptophan Trp content: phosphorylase B PB, 1. Gels were run for 20 min at V and imaged using a stain-free enabled imager left. Gels were then stained with Coomassie stain and imaged using the same imager right. Purification of recombinant proteins is a critical step during protein therapeutics development. Protein therapeutics have a number of classifications based on their potential applications, including use as vaccines and diagnostics as well as for enzymatic, regulatory, or targeting activities 1.

Chromatography is an important biophysical technique that enables the separation, identification, and purification of the components of a mixture for qualitative and quantitative analysis. Proteins can be purified based on characteristics such as size and shape, total charge, hydrophobic groups present on the surface, and binding capacity with the stationary phase.

Protein Purification: Principles and Practice
